目的探讨盐酸吉西他滨对乳腺癌小鼠肿瘤抑制因子候选基因2(Tusc2)、大型肿瘤抑制因子2(Lats2)基因mRNA及蛋白水平表达的影响。方法首先建立小鼠乳腺癌模型,盐酸吉西他滨药物处理前(0d)、处理后第5天(5d)、处理后第10天(10d)分别取材,采用RT-PCR和Western blotting方法检测Tusc2、Lats2基因表达量的变化。结果盐酸吉西他滨作用乳腺癌小鼠第10天(10d)后,RT-PCR结果显示,与空白对照组相比,实验组Tusc2与Lats2 mRNA相对表达量分别增加约4.5倍与8倍(P<0.05),其余各组与相应空白对照组相比,相对表达量无统计学差异(P>0.05);同时Western blotting结果显示,实验组与空白对照组相比,TUSC2与LATS2蛋白相对表达量分别上调约10倍与5倍(P<0.05)。其他各组之间相比统计学无显著差异(P>0.05)。结论盐酸吉西他滨能有效上调乳腺癌小鼠体内Tusc2、Lats2基因的表达。
Objective To investigate the effect of gemcitabine hydrochloride on the expression of Tusc2 and Lats2 mRNA and protein in breast cancer mice. Methods The mouse models of breast cancer, gemcitabine hydrochloride pretreatment (0d), the fifth day (5d) and the tenth day (10d) of treatment were established. The expression of Tusc2, Lats2 Changes in gene expression. Results The results of RT-PCR showed that the relative expression of Tusc2 and Lats2 mRNA in experimental group increased by about 4.5-fold and 8-fold (P <0.05) compared with that of the blank control group on the 10th day after gemcitabine hydrochloride treatment (P> 0.05). The results of Western blotting showed that the relative expression of TUSC2 and LATS2 protein in the experimental group was significantly higher than that in the blank control group About 10 times and 5 times (P <0.05). There was no significant difference between the other groups (P> 0.05). Conclusion Gemcitabine hydrochloride can effectively up-regulate Tusc2 and Lats2 gene expression in breast cancer mice.
