目的观察解毒通络方含药血清对大鼠血管成纤维细胞释放内皮素(ET)、一氧化氮(NO)、基质金属蛋白酶-9(MMP-9)的影响。方法体外培养大鼠血管成纤维细胞,以血管紧张素Ⅱ(AngⅡ)建立血管成纤维细胞增殖模型,通过酶联免疫吸附法观察含药血清对其释放ET、NO、和MMP-9的影响。结果解毒通络方含药血清各个剂量组能抑制血管成纤维细胞ET、MMP-9释放,提高血管成纤维细胞NO释放。结论解毒通络方含药血清能够调节AngⅡ诱导增殖血管成纤维细胞释放ET、NO和MMP-9的含量。
Objective To observe the effect of Jiedu Tongluo containing serum on the release of endothelin (ET), nitric oxide (NO) and matrix metalloproteinase-9 (MMP-9) from rat vascular fibroblasts. Methods Rat vascular fibroblasts were cultured in vitro. The proliferation model of vascular fibroblasts was established by angiotensin Ⅱ (Ang Ⅱ). The effect of containing serum on ET, NO and MMP-9 release was observed by enzyme-linked immunosorbent assay. Results Jiedu Tongluo containing serum in each dose group can inhibit the release of ET and MMP-9 in vascular fibroblasts and increase the release of NO in vascular fibroblasts. Conclusion Jiedu Tongluo containing serum can regulate the content of ET, NO and MMP-9 released from Ang II-induced proliferation of vascular fibroblasts.
