PDGFR-β基因敲除小鼠上丘内Otx2和PV免疫反应阳性神经元的分布

目的:探讨血小板源性生长因子受体β(platelet-derived growth factor receptor-β,PDGFR-β)在上丘神经元的作用。方法:采用免疫细胞化学法和图像分析技术观察正常成年小鼠和PDGFR-β基因敲除小鼠上丘内Otx2和小白蛋白(parvalbumin,PV)免疫反应阳性神经元的形态、数目、大小及分布。结果:PDGFR-β基因敲除小鼠上丘内Otx2和PV免疫反应阳性神经元的基本形态及分布特征与正常小鼠无明显差别,即Otx2免疫反应阳性神经元密集地分布于上丘的浅层,而PV免疫反应阳性神经元则分布于上丘的各层。但这两种神经元的数目在PDGFR-β基因敲除小鼠却明显减少,其平均直径也较正常小鼠者明显为小。结论:PDGFR-β可能通过Otx2和PV等神经化学物质,参与上丘的功能形成与发育。 Objective: To investigate the effect of platelet-derived growth factor receptor-β (PDGFR-β) on the superior colliculus neurons. Methods: Immunocytochemistry and image analysis were used to observe the morphology, number, size and expression of Otx2 and parvalbumin (PV) immunoreactive neurons in the superior colliculus in normal adult mice and PDGFR-β knockout mice distributed. Results: The basic morphology and distribution of Otx2 and PV immunoreactive neurons in the superior colliculus of PDGFR-β knockout mice were similar to those of normal mice. Otx2 immunoreactive neurons were densely distributed in the superior colliculus Layer, and PV immunoreactive neurons are distributed in the upper layers of the hill. But the number of these two neurons in PDGFR-β knockout mice was significantly reduced, the average diameter is also significantly smaller than normal mice. Conclusion: PDGFR-β may be involved in the functional formation and development of the superior colliculus through neurochemicals such as Otx2 and PV.
