目的观察新生小鼠视网膜发育过程中单核细胞趋化蛋白-1(MCP-1)的表达,探讨其在视网膜血管发育过程中的作用。方法选择C57BL/6J幼鼠(分别为3、5、7、12、14、21日龄各10只)60只。将所有小鼠按日龄处死,立即摘除眼球,取视网膜组织,分别采用RT-PCR、Western blot和免疫组化染色检测MCP-1 mRNA和蛋白。结果 MCP-1 mRNA和蛋白在3日龄小鼠视网膜组织中即有表达,12日龄时呈现一过性高表达,之后迅速下降,21日龄时表达基本呈基线水平。结论小鼠的视网膜发育过程始终伴随MCP-1的表达,其在12日龄时(视网膜血管发育关键期)的高表达可能与视网膜血管的发育和迅速成熟有关。
Objective To observe the expression of monocyte chemoattractant protein-1 (MCP-1) during retina development in neonatal mice and to explore its role in retinal vascular development. Methods Sixty C57BL / 6J young rats (10, 10, respectively, 3, 5, 7, 12, 14 and 21 days old) were selected. All mice were sacrificed at the age of day. The eyeballs were immediately removed and the retina was removed. The mRNA and protein of MCP-1 were detected by RT-PCR, Western blot and immunohistochemistry respectively. Results The mRNA and protein of MCP-1 were expressed in the retina of 3-day-old mice. The expression of MCP-1 mRNA was transiently elevated on the 12th day of age and then decreased rapidly. At 21 days of age, the expression of MCP-1 mRNA and protein was basically at the baseline level. Conclusion The development of retina in mice is always accompanied by the expression of MCP-1. The high expression of MCP-1 at 12 days of age (the critical stage of retinal vascularization) may be related to the development and rapid maturation of retinal vessels.
