目的:设计微流控芯片,单细胞分析慢性粒细胞白血病患者粒细胞表达髓过氧化物酶(MPO)活性。方法:收集慢性粒细胞白血病患者和对照组各60例,联苯胺-H2O2法对血细胞进行MPO染色。应用自行设计的微流控芯片单细胞分析粒细胞表达MPO。结果:联苯胺染色法发现粒细胞中表达MPO,且各阶段染色程度不同,MPO呈现颗粒状分布。芯片分析的结果与联苯胺染色法所呈现的结果在形态特征方面一致,试验组和对照组成熟粒细胞MPO活性均高于幼稚粒细胞,平均MPO活性试验组明显高于对照组。结论:毛细管芯片单细胞分析粒细胞MPO活性,发现慢性粒细胞白血病患者粒细胞平均MPO显著增高,有可能作为慢性粒细胞白血病诊断标志和个体化检查指标。
OBJECTIVE: To design a microfluidic chip for single cell analysis of myeloperoxidase (MPO) activity in granulocytes of patients with chronic myeloid leukemia. Methods: 60 patients with chronic myeloid leukemia and control group were collected, MPO staining was performed on blood cells by benzidine-H2O2 method. Single cell analysis of granulocytes using self - designed microfluidic chip for MPO expression. Results: The benzidine staining showed that MPO was expressed in granulocytes, and the degree of staining was different in each stage. The MPO showed granular distribution. The result of chip analysis was consistent with that of benzidine staining in morphological characteristics. The MPO activity of mature granulocytes in experimental group and control group was higher than that in immature granulocytes, and the average MPO activity in experimental group was significantly higher than that in control group. Conclusion: The single cell of capillary chip analysis of MPO activity of granulocytes, found that patients with chronic myeloid leukemia mean MPO was significantly higher, it may be used as a diagnostic marker of chronic myeloid leukemia and individualized indicators.
