目的:研究盆腔炎颗粒对慢性盆腔炎大鼠核因子κB(NF-κB)及相关指标表达的影响。方法:以混合细菌加机械损伤法复制大鼠慢性盆腔炎模型,随机分为正常对照组、假手术组、模型组、盆炎净组及盆腔炎颗粒大、中、小剂量组,造模2周后,各治疗组灌胃给药2周,免疫组化法检测各组大鼠子宫组织中NF-κB、核因子κB结合蛋白(IκB)、细胞间黏附分子-1(ICAM-1)、肿瘤坏死因子-α(TNF-α)的表达,逆转录-聚合酶链反应法检测各组大鼠子宫组织中ICAM-1mRNA、TNF-αmRNA表达。结果:盆腔炎颗粒可以显著降低模型大鼠NF-κB、IκB、TNF-α、ICAM-1、TNF-αmRNA及ICAM-1mRNA的表达,盆腔炎中剂量组子宫组织NF-κB与TNF-αmRNA及ICAM-1mRNA表达之间均呈显著正相关。结论:NF-κB活化的抑制在活血补肾法治疗慢性盆腔炎抗炎及抗粘连机制中起重要作用。
Objective: To study the effect of pelvic inflammatory granules on the expression of nuclear factor-κB (NF-κB) and related indicators in rats with chronic pelvic inflammatory disease. Methods: Chronic pelvic inflammatory disease rats were induced by mixed bacteria and mechanical injury and were randomly divided into normal control group, sham operation group, model group, pelvic inflammatory disease group, pelvic inflammatory disease group, model 2 The rats in each group were given gavage for 2 weeks. The expression of NF-κB, nuclear factor κB-binding protein (IκB), intercellular adhesion molecule-1 (ICAM-1) Tumor necrosis factor-α (TNF-α) was detected by enzyme-linked immunosorbent assay (RT-PCR). The expression of ICAM-1mRNA and TNF-αmRNA in uterus of rats in each group were detected by reverse transcription polymerase chain reaction. Results: Pelvic inflammatory disease particles can significantly reduce the expression of NF-κB, IκB, TNF-α, ICAM-1, TNF-αmRNA and ICAM-1mRNA in the model rats, There was a significant positive correlation between ICAM-1mRNA expression and ICAM-1mRNA expression. Conclusion: The inhibition of NF-κB activation plays an important role in the treatment of chronic pelvic inflammatory disease and anti-adhesion mechanism.
