目的了解余姚市鼠形动物汉坦病毒(HV)携带情况及基因型别。方法选择余姚市的平原、山区、滨海和城区四种地形,采用夹夜法捕获鼠类动物248只,采用实时荧光定量PCR法检测鼠类动物肺标本HV核酸,采用反转录PCR法扩增HV核酸阳性样本的M基因并进行测序和分析。结果共检出HV核酸阳性样本12份,分布于除城区外的其余3个地区,总带毒率为4.84%,且均在褐家鼠中检出;构建系统发生树分析其中8份样本的核苷酸序列,结果显示其M基因与Bj HD01核苷酸同源性最高为98.3%~99.8%,与浙江株Z37和ZT10核苷酸同源性为96.2%~97.6%,均为汉城病毒(SEOV)S3亚型。结论余姚市鼠类动物中携带的HV阳性检出率较高,褐家鼠为主要带毒宿主,基因亚型分布较单一,以SEOV S3亚型为主。
Objective To understand the carrying status and genotype of Hantaan virus (HV) in Yuyao rat. Methods Four terrains of plain, mountainous area, coastal area and urban area were selected in Yuyao City. 248 animals were captured by the method of nocturnent night. The HV nucleic acids of the lung specimens were detected by real - time fluorescence quantitative polymerase chain reaction (RT - PCR) M nucleic acid positive samples were sequenced and analyzed. RESULTS: Twelve HV positive samples were detected in the remaining three districts except for urban areas, with a total virulence of 4.84% and were detected in Rattus norvegicus. Eight of the eight samples were constructed by phylogenetic tree analysis The results showed that the homology of M gene with Bj HD01 nucleotide was 98.3% ~ 99.8% and that of Zhejiang strain Z37 and ZT10 was 96.2% ~ 97.6%, both of which were Seoul virus (SEOV) S3 subtype. Conclusion The detection rate of HV positive in rodents in Yuyao is relatively high. Rattus norvegicus is the main virulent host. The genotype distribution of the genotypes is single, with the majority of SEOV S3 subtypes.
