旨在研究肉鸡腹水综合征发生发展过程中的肺组织EGFL7、VEGF和VEGFR2等血管内皮细胞源性增殖因子表达量的变化和复方中药对其影响。275羽8日龄罗斯肉鸡随机分为空白组(常规饲养),模型组(9~11℃,饲料中添加3%猪油和4%鱼粉,饮水中添加0.12%NaCl),复方中药高、中、低剂量组(饲养同模型组,每天于饮水中分别给予2、1、0.5mL·kg-1复方中药),用荧光定量PCR和ELISA检测15、25、35、45日龄肉鸡肺组织EGFL7、VEGF、VEGFR2的基因和蛋白质表达量。结果表明:与空白组相比,模型组肉鸡腹水心脏指数极显著升高(P<0.01),肺组织EGFL7、VEGF、VEGFR2的基因和蛋白质表达量显著增加(P<0.01或P<0.05);与模型组相比,复方中药高、中剂量组均能不同程度地改善肉鸡的精神状态、腹水、心包积液等症状,能显著下调腹水心脏指数和肺组织EGFL7、VEGF、VEGFR2的基因和蛋白质表达量(P<0.01或P<0.05)。腹水综合征肉鸡肺组织EGFL7、VEGF、VEGFR2的基因和蛋白质表达量升高,参与了肉鸡腹水综合征病理的发生发展;复方中药能下调其表达,保护肺组织血管内皮功能,有效防治肉鸡腹水综合征。
The purpose of this study is to investigate the changes of expression of EGFR7, VEGF and VEGFR2 in the development of broiler ascites syndrome and the effect of compound Chinese herbs on it. 275 eight-day old Rose broilers were randomly divided into blank group (conventional feeding), model group (9-11 ℃, 3% lard and 4% fish meal in feed, 0.12% NaCl in drinking water) , Low dose group (fed with the model group, respectively, in the drinking water were given 2,1,0.5mL · kg-1 compound Chinese medicine), fluorescence quantitative PCR and ELISA detection of 15, 25, 35, 45 days old broiler lung tissue EGFL7 , VEGF, VEGFR2 gene and protein expression levels.
