目的以原代培养小鼠骨髓基质细胞为饲养层,探讨精原干细胞能否在该饲养层上生长及生长的影响因素。方法采用原代培养的小鼠骨髓基质细胞,待细胞长成单层后用丝裂霉素C处理并接种生精细胞共培养。结果原代的小鼠培养骨髓基质细胞能很好的维持精原干细胞的非分化性扩增。结论小鼠原代培养的骨髓基质细胞能作为饲养层支持精原干细胞体外非分化性扩增,该细胞有望成为干细胞培养的新的饲养层。
Objective To culture primary mouse bone marrow stromal cells as a feeder layer to investigate whether spermatogonial stem cells can grow and grow on this feeder layer. Methods Primary cultured mouse bone marrow stromal cells were used. After the cells grew into monolayer, they were treated with mitomycin C and inoculated with spermatogenic cells. The results of primary mouse bone marrow stromal cells cultured well maintained undifferentiated spermatogonial expansion. Conclusion The mouse primary culture of bone marrow stromal cells can support spermatogonial stem cell proliferation in vitro as feeder layer, which is expected to become a new feeder layer for stem cell culture.
