目的:研究二色波罗蜜根皮总黄酮的最佳提取、纯化工艺条件。方法:采用正交试验设计,以总黄酮含量为考察指标研究二色波罗蜜根皮总黄酮的最佳提取工艺条件;以总黄酮的静态吸附量和解吸率为考察指标筛选大孔吸附树脂,并优化总黄酮的纯化工艺条件。结果:最优提取工艺条件为乙醇浓度为60%,浸提温度为50℃,浸提次数为1次,料液比为1∶8。HPD-500型大孔吸附树脂对二色波罗蜜根皮总黄酮有较好的吸附分离性能,以22.0mg总黄酮/g树脂的上柱量配制成50.0 mg/mL的上柱液,上HPD-500大孔吸附树脂柱,静置1 h,用4 BV的水洗脱杂质,再用4 BV的80%乙醇洗脱,得到的精制品中总黄酮含量为86.4%,树脂富集倍数为5.33。结论:采用以上提取、纯化条件进行二色波罗蜜根皮总黄酮的制备,简便、实用,可以为工业生产提供参考。
Objective: To study the optimum extraction and purification of total flavonoids from Apocynum venetum. Methods: The optimum extraction conditions of total flavonoids of Rhizoma Dioscoreae were studied based on the total flavonoids content by orthogonal experimental design. The macroporous resin was screened by the static adsorption capacity and desorption rate of total flavonoids Optimization of the purification of total flavonoids process conditions. Results: The optimum extraction conditions were ethanol concentration of 60%, extraction temperature of 50 ℃, extraction times of 1 and solid-liquid ratio of 1: 8. HPD-500 macroporous resin has good adsorption and separation performance on the total flavonoids of Apocynum bark, with the amount of 22.0mg total flavonoids / g resin on the column preparation of 50.0 mg / mL on the HPD- 500 macroporous resin column was allowed to stand for 1 h. The impurities were eluted with 4 BV of water and then eluted with 4 BV of 80% ethanol. The resulting purified product had a total flavonoid content of 86.4% and a resin enrichment factor of 5.33 . Conclusion: The above extraction and purification conditions for the preparation of the total flavonoids of Apocynum bisporus, simple and practical, can provide a reference for industrial production.
