目的研究基质金属蛋白酶2(MMP-2)、尿激酶型纤溶酶原激活物(uPA)的表达情况以及它们与结肠癌的关系。方法收集了55例病理学确证的结肠癌组织标本(病例组)和30例结肠癌癌旁的正常组织标本(对照组),采用免疫组织化学方法(SP法)检测其MMP-2、uPA的表达。结果病例组中的MMP-2、uPA的表达率分别为74.54%、61.82%,对照组的MMP-2、uPA的表达率分别为33.33%、16.67%,2组比较差异均有统计学意义(MMP-2,χ2=13.737,P<0.001;uPA,χ2=15.938,P<0.001)。结肠癌组织中MMP-2、uPA的表达与癌组织的浸润深度、淋巴结转移、肿瘤分期显著相关(P均<0.05)。MMP-2与uPA的表达呈正相关(rs=0.572,P=0.000)。结论MMP-2、uPA的表达与结肠癌的侵袭、转移密切相关,二者联合检测可作为判断结肠癌发展及预后的分子指标。
Objective To investigate the expression of matrix metalloproteinase 2 (MMP-2) and urokinase-type plasminogen activator (uPA) and their relationship with colon cancer. Methods Collected 55 cases of pathologically confirmed colon cancer tissue samples (case group) and 30 cases of colon cancer adjacent normal tissue samples (control group), immunohistochemistry (SP method) detection of MMP-2, uPA expression. Results The expression rates of MMP-2 and uPA in the case group were 74.54% and 61.82%, respectively. The expression rates of MMP-2 and uPA in the control group were 33.33% and 16.67% respectively. There were significant differences between the two groups MMP-2, χ2 = 13.737, P <0.001; uPA, χ2 = 15.938, P <0.001). The expressions of MMP-2 and uPA in colon cancer tissues were significantly correlated with the depth of invasion, lymph node metastasis and tumor stage (all P <0.05). The expression of MMP-2 and uPA was positively correlated (rs = 0.572, P = 0.000). Conclusion The expression of MMP-2 and uPA is closely related to the invasion and metastasis of colon cancer. The combined detection of MMP-2 and uPA may be used as a molecular index to judge the development and prognosis of colon cancer.
