目的观察益肺活血颗粒对慢性肺源性心脏病(CPHD)家兔相关血管细胞因子血管紧张素Ⅱ(AngⅡ)、血管内皮素1(ET-1)、肿瘤坏死因子α(TNF-α)、一氧化氮(NO)以及6酮前列腺素F1a(6-Keto-PGF1a)的影响,探讨其改善肺动脉高压的作用机制。方法采用0.7%FeCl3溶液耳缘静脉注射造模方法建立CPHD家兔模型(隔日注射1次,连续21次,共42 d,总剂量约15 mL/只),随机设立正常组、模型组、卡托普利组及益肺活血颗粒组。放射免疫法检测AngⅡ、ET-1、TNF-α和6-Keto-PGF 1a,硝酸还原酶法测定NO。结果 42 d时,模型组与正常组比较,AngⅡ、ET-1、TNF-α含量明显升高,二者有显著性差异(P<0.05);6-Keto-PGF1a、NO含量明显降低,二者有显著性差异(P<0.01)。62 d时,益气活血颗粒组与模型组比较,Ang-Ⅱ、ET-1含量明显降低(P<0.05),NO含量升高(P<0.05)。结论益气活血颗粒通过降低AngⅡ、ET-1含量,升高NO含量,舒张肺动脉血管、降低血管阻力,防治慢性肺源性心脏病。
Objective To observe the effects of Yifeuhuoxue Granule on angiotensin Ⅱ (AngⅡ), endothelin-1 (ET-1), tumor necrosis factor-α (TNF-α) Nitric oxide (NO) and 6-keto-prostaglandin F1a (6-Keto-PGF1a), and to explore its mechanism of improving pulmonary hypertension. Methods CPHD rabbits were established by ear vein injection of 0.7% FeCl3 solution (injected once every other day for 21 consecutive days for a total of 42 days with a total dose of about 15 mL / mouse). Normal rabbits were randomly divided into normal group, model group, Topril group and Yifeihuoxue granule group. The levels of AngⅡ, ET-1, TNF-α and 6-Keto-PGF 1a were detected by radioimmunoassay and NO was determined by nitrate reductase method. Results At 42 days, the levels of AngⅡ, ET-1 and TNF-α in the model group were significantly higher than those in the normal group (P <0.05), and the contents of 6-Keto-PGF1a and NO were significantly decreased Who had significant difference (P <0.01). At 62 days, the content of Ang-Ⅱ and ET-1 in the Yiqi Huoxue granule group was significantly lower than that in the model group (P <0.05), and the NO content was increased (P <0.05). Conclusion Yiqi Huoxue Granule can prevent and treat chronic cor pulmonale by decreasing the content of AngⅡ and ET-1, increasing the content of NO, relaxing the pulmonary artery, decreasing the vascular resistance.
