用地高辛标记的HBV基因组探针检测了66例肝细胞性肝癌(HCC)中HBV的存在状态,从中筛选出32例仅有HBVDNA整合的HCC作为研究对象,进一步检测了HBVX基因、S基因、Pre-S、C基因DNA片段的整合情况,并探讨了HBV4种基因亚片段整合与ras、myc癌基因表达的关系。结果显示,HBVX、S、Pre-S和C基因片段的整合率分别为87.5%(28/32)、62.5%(20/32)、62.5%(20/32)和25.0%(8/32)。p21ras、p62myc在HCC中的表达率分别为50.0%(20/40)、81.2%(26/32)。通过对HCC中4种HBV基因片段整合与p21ras、p62myc表达之间关系的研究,显示p62myc的表达与HBVX、Pre-S基因整合关系密切,p21ras的表达则主要与HBVX基因整合有关(P<0.05)。提示HBVX、Pre-SDNA片段在宿主细胞染色体上的整合可能直接或通过其蛋白启动myc和(或)ras癌基因的表达。
