目的本实验研究长期低剂量激素替代疗法(HRT)对绝经后妇女体内激素水平及外周型苯二氮卓受体(PBR)的影响。方法筛选北京协和医院医护人员中符合入组条件并接受HRT治疗持续4年以上志愿者83人,同时以年龄为配比条件筛选出99人作为对照。以酶联免疫反应法测血浆雌二醇、孕酮、睾酮含量;以同位素标记法测外周血PBR结合活性。结果与对照组相比,HRT组受试者血浆中雌二醇维持在较高水平,两组之间在各个年龄段都有显著差异(P<0.05),而血浆中孕酮、睾酮水平未见显著差异。各年龄段HRT组PBR水平均高于对照组,并在50~59年龄段两组之间有显著性差异(P<0.01)。受试者血中PBR结合活性与雌激素水平呈明显相关性。结论长期低剂量HRT使绝经后受试者体内雌激素水平维持在一个较高浓度的同时血小板上PBR亦处于较高的水平。
Objective This study was to investigate the effect of long-term low-dose hormone replacement therapy (HRT) on hormone levels and peripheral benzodiazepine receptor (PBR) in postmenopausal women. Methods Screening of Beijing Union Medical College Hospital staff who meet the inclusion criteria and who received HRT for more than 4 years, 83 volunteers, while 99 were selected as the control by the age-matched conditions. Serum estradiol, progesterone and testosterone levels were measured by enzyme-linked immunosorbent assay. Peripheral blood PBR binding activity was measured by isotope labeling. Results Compared with the control group, the level of estradiol in plasma of HRT group was maintained at a high level, and there was significant difference between the two groups in all age groups (P <0.05), while the level of plasma progesterone and testosterone See significant differences. PBR levels in HRT group were higher than those in control group at all ages, and there was significant difference between the two groups in 50 ~ 59 (P <0.01). PBR binding activity in the blood of the subjects was significantly correlated with estrogen levels. Conclusions Long-term, low-dose HRT maintains estrogen levels in postmenopausal subjects at a higher concentration and higher PBR on platelets.
