目的:HPLC梯度洗脱法同时测定天山花楸叶提取物中的芦丁、金丝桃苷、异槲皮苷、槲皮素-3-O-(6″-O-丙二酰基)-β-D-葡萄糖苷、紫云英苷、山柰素-3-O-(6″-O-丙二酰基)-β-D-葡萄糖苷。方法:采用Waters C18色谱柱(4.6 mm×150 mm,5μm),乙腈-0.4%磷酸溶液为流动相梯度洗脱,运行时间30.0 min,平衡时间10 min,流速1.0 mL·min-1,柱温25℃,波长360 nm。结果:芦丁、金丝桃苷、异槲皮苷、槲皮素-3-O-(6″-O-丙二酰基)-β-D-葡萄糖苷、紫云英苷、山柰素-3-O-(6″-O-丙二酰基)-β-D-葡萄糖苷分别在3.3~39.5,6.2~74.1,12.3~147.5,14.3~171.2,7.5~90.4,15.6~187.5 mg·L-1与峰面积呈良好的线性关系;平均加样回收率分别为98.1%,97.5%,94.5%,95.8%,102.2%,97.9%,其RSD分别为1.37%,1.57%,1.02%,1.77%,2.48%,1.38%。结论:该法操作简便、快捷、准确、稳定性及重复性好,可用于对天山花楸中6个成分的含量测定。
OBJECTIVE: To determine the contents of rutin, hyperoside, isoquercitrin, quercetin -3-O- (6 “-O-malonyl) -β -D-glucoside, alisin, behen-3-O- (6 ”-O-malonyl) -beta-D-glucoside. Methods: The mobile phase was eluted with a gradient of acetonitrile-0.4% phosphoric acid on a Waters C18 column (4.6 mm × 150 mm, 5 μm) with a running time of 30.0 min, an equilibration time of 10 min and a flow rate of 1.0 mL · min- 25 ℃, wavelength 360 nm. RESULTS: Rutin, hyperoside, isoquercetin, quercetin -3-O- (6 “-O-malonyl) -β-D-glucoside, 3-O- (6 ”-O-malonyl) -β-D-glucoside were detected in the range of 3.3 ~ 39.5,6.2 ~ 74.1,12.3 ~ 147.5,14.3 ~ 171.2,7.5 ~ 90.4,15.6 ~ 187.5 mg · L- 1 and peak area showed a good linear relationship. The average recoveries were 98.1%, 97.5%, 94.5%, 95.8%, 102.2% and 97.9%, respectively. The RSDs were 1.37%, 1.57%, 1.02% and 1.77% , 2.48%, 1.38%. Conclusion: The method is simple, rapid, accurate, stable and reproducible. It can be used for the determination of six components in the Sorbus tianschanica.
