目的研究SIRS模型肠组织中TFmRNA的表达,探讨TF在SIRS发生中的作用。方法 40只新生Wistar大鼠,出生36h~48h,随机分成正常对照组(A组)10只及模型组(B组)30只。48小时后处死动物,根据血液白细胞总数及呼吸改变确定为SIRS。将B组分为未发生SIRS组(B1组)和发生SIRS组(B2组)。实时荧光定量聚合酶链反应(QRT-PCR)检测肠组织TFmRNA的表达。结果肠组织TFmRNA表达水平(2-ΔΔCt值)∶A组1.01±0.04,B1组2.14±0.19,B2组3.78±1.2,三组间比较差异有显著性(P<0.05)。结论从转录水平证实,SIRS新生大鼠模型中,TF水平增加,提示TF参与炎症反应。
Objective To study the expression of TF mRNA in intestinal tissue of SIRS model and to explore the role of TF in the pathogenesis of SIRS. Methods Forty newborn Wistar rats were born from 36h to 48h. They were randomly divided into 10 normal control group (A group) and 30 model group (B group). Animals were sacrificed 48 hours later and were identified as SIRS based on the total number of leukocytes and respiratory changes. Group B was divided into the group without SIRS (group B1) and the group with SIRS (group B2). Real-time fluorescent quantitative polymerase chain reaction (QRT-PCR) was used to detect the expression of TFmRNA in intestinal tissue. Results The expression level of TF mRNA (2-ΔΔCt value) in intestinal tissue was 1.01 ± 0.04 in group A, 2.14 ± 0.19 in group B1 and 3.78 ± 1.2 in group B2, respectively. There was significant difference among the three groups (P <0.05). Conclusion From the transcriptional level, it is confirmed that the level of TF is increased in the newborn rat model of SIRS, suggesting that TF participates in the inflammatory reaction.
