目的建立高效毛细管电泳法快速检测猪肉和内脏中克伦特罗、沙丁胺醇和特布他林含量的方法。方法匀浆样品经丙酮溶液超声波萃取,萃取液经离心和过滤后用氮气吹干。用磷酸盐缓冲液溶解残渣,低温离心,取上清过滤后进样。电泳条件:石英毛细管(75/365μm,40/47 cm),缓冲液为2μg/ml季铵化纤维素负载的纳米金磷酸盐缓冲液(p H=10.5),电压为12 k V,检测波长为210 nm。结果 3种物质均在5 min内出峰。浓度为1μg/ml~250μg/ml时,3种物质的峰面积和浓度的线性关系良好,相关系数r分别为0.996 7、0.994 8和0.995 3。猪肉和猪肝中3种物质的加标回收率为87%~113%,精密度RSD为2.8%~6.5%。结论该方法具有快速、简便、准确度和精密度高等特点,适用于猪肉和猪肝中克伦特罗、沙丁胺醇和特布他林的快速检测。
OBJECTIVE To establish a rapid method for the determination of clenbuterol, salbutamol and terbutaline in pork and offal by high performance capillary electrophoresis. Methods The homogenized sample was extracted with acetone by ultrasonic extraction. The extract was centrifuged and filtered, and then blown dry with nitrogen. Resolve the residue with phosphate buffer, centrifuged at low temperature, the supernatant filtered and injected. Electrophoresis conditions: quartz capillary (75/365 μm, 40/47 cm), buffer 2 μg / ml quaternized cellulose loaded gold gold phosphate buffer (p H = 10.5), voltage 12 kV, detection wavelength 210 nm. Results All the three substances peaked within 5 min. The linear relationship between the peak area and the concentration of the three substances was good at the concentration of 1μg / ml ~ 250μg / ml with the correlation coefficients r of 0.996 7, 0.994 8 and 0.995 3, respectively. The spiked recoveries of the three substances in pork and pig liver ranged from 87% to 113% with RSDs of 2.8% to 6.5%. Conclusion The method is rapid, simple, accurate and precise. It is suitable for the rapid detection of clenbuterol, salbutamol and terbutaline in pork and pork liver.
