目的 探讨理冲消瘤汤对子宫肌瘤大鼠模型凋亡基因的调控作用.方法 制备子宫肌瘤大鼠模型,随机分为正常组、模型组、理冲消瘤汤高剂量组、理冲消瘤汤低剂量组、阳性对照组,灌胃给药,连续4周.电镜观察子宫肌瘤细胞微观结构;采用实时荧光定量PCR (Real-time PCR)检测细胞凋亡调控基因(Survivin、Caspase-3)的表达;采用免疫荧光检测细胞凋亡调控基因(Survivin、Caspase-3)的表达和分布.结果 与阳性对照组比较,理冲消瘤汤组细胞排列基本正常,细胞器基本接近正常,线粒体增生肥大减轻,胶原纤维排列基本规则,增生不明显;Real-time PCR结果显示,理冲消瘤汤组的Survivin表达降低(P<0.01),Caspase-3的表达升高(P<0.05);免疫荧光结果显示,与阳性对照组比较,理冲消瘤汤组的Survivin表达显著降低(P<0.05),而Caspase-3的表达显著升高(P<0.05).结论 理冲消瘤汤对子宫肌瘤细胞的生长有抑制作用,可改善子宫肌纤维紊乱和肌瘤细胞的超微结构.其作用机制可能与其调控子宫肌瘤平滑肌中凋亡基因Survivin、Caspase-3的表达有关.“,”Objective To explore the regulation effect of Lichong Xiaoliu decoction on apoptosis gene of rats with uterine fibroids.Methods The rat model of uterine fibroids was established,then the rats were divided into normal group,model group,high-dose Lichong Xiaoliu decoction group,low-dose Lichong Xiaoliu decoction group,and positive control group,the rats were treated by intragastric administration for consecutive four weeks.Electron microscope was used to observe microstructure of uterine fibroid cells.Real-time fluorescent quantitative PCR was used to detect the expressions of Survivin and Caspase-3.Immunofluorescence was used to detect the expressions and distributions of Survivin and Caspase-3.Results Compared with positive control group,cell arrangement in Lichong Xiaoliu decoction group was basically normal,organelles were normal,hypertrophy of mitochondria was relieved,arrangement of collagenous fibers was regular,hyperplasia was not obvious.Real-time fluorescent quantitative PCR showed that the expression of Survivin in Lichong Xiaoliu decoction group decreased significantly (P < 0.01),while the expression of Caspase-3 increased significantly (P < 0.05).Immunofluorescence showed that compared with positive control group,the expression of Survivin in Lichong Xiaoliu decoction group decreased significantly (P< 0.05),while the expression of Caspase-3 increased significantly (P<0.05).Conclusion Lichong Xiaoliu decoction can inhibit the growth of uterine fibroid cells and improve uterine myofibrillar disarray and microstructure of uterine fibroid cells.The mechanism may be correlated with regulation effect of Lichong Xiaoliu decoction on expressions of Survivin and Caspase-3 in uterine fibroid cells.
