在THP-1细胞中,研究依博素对炎症细胞因子白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)及肿瘤坏死因子α(TNFα)的作用,以探讨其抗炎作用机制。采用荧光标记底物,证明了依博素对IL-1β转化酶(ICE)和TNFα转化酶(TACE)活性的抑制作用;应用实时定量PCR(real-time PCR)确定了依博素对上述炎症细胞因子mRNA表达水平的降低作用;利用酶联免疫吸附方法 (ELISA)及Western blotting检测证明,依博素对3种炎症细胞因子分泌水平的抑制作用。上述结果表明,在THP-1细胞中,依博素可通过抑制ICE酶和TACE酶活性、降低炎症细胞因子mRNA合成作用而使各炎症细胞因子的生成量下降。本研究在细胞水平初步考察了依博素的抗炎作用机制,为治疗类风湿性关节炎新药研究奠定了相关基础。
In THP-1 cells, the effect of Ebosin on inflammatory cytokines interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor α (TNFα) Anti-inflammatory mechanism. Using fluorescent labeled substrate, the inhibitory effect of ebosin on IL-1βconverting enzyme (ICE) and TNFα converting enzyme (TACE) activity was demonstrated. The effect of ebosin on the above inflammation was confirmed by real-time PCR Cytokines mRNA expression levels decreased; enzyme-linked immunosorbent assay (ELISA) and Western blotting test showed that Ebosu on the secretion of three kinds of inflammatory cytokines levels. The above results show that in THP-1 cells, ebosin can reduce the production of various inflammatory cytokines by inhibiting ICE enzyme and TACE enzyme activity and reducing the mRNA synthesis of inflammatory cytokines. This study initially investigated the anti-inflammatory mechanism of Ebosin at the cellular level, which laid the foundation for the research of new drugs for the treatment of rheumatoid arthritis.
