目的探讨氯化镧对人脑胶质瘤SHG44细胞增殖的影响及其作用机制。方法体外培养人胶质瘤细胞,MTT法测定氯化镧对SHG44细胞的增殖影响,电镜检查细胞超微结构的改变,原位杂交技术及免疫组织化学染色技术检测PTEN基因及其蛋白表达情况。结果氯化镧可抑制人脑胶质瘤SHG44细胞的生长,电镜下见胶质瘤细胞核染色质浓缩、核固缩、核溶解,出现凋亡细胞。PTEN基因在蛋白、mRNA水平均显著增强。结论氯化镧可抑制人脑胶质瘤SHG44细胞的生长,并促进凋亡。
Objective To investigate the effect of lanthanum chloride on the proliferation of human glioma SHG44 cells and its mechanism. Methods The human glioma cells were cultured in vitro. The effects of lanthanum chloride on the proliferation of SHG44 cells were determined by MTT assay. The ultrastructure of cells was examined by electron microscopy. The expression of PTEN gene and its protein were detected by in situ hybridization and immunohistochemical staining. Results Lanthanum chloride could inhibit the growth of human glioma SHG44 cells. Under the electron microscope, the chromatin condensation, nuclear pyknosis and nuclear lysis of glioma cells were observed. Apoptotic cells appeared. PTEN gene in the protein and mRNA levels were significantly enhanced. Conclusion Lanthanum chloride can inhibit the growth of human glioma SHG44 cells and promote apoptosis.
