目的 :寻找适合临床实际需要的逆转多药耐药的方法。方法 :用 MTT法和流式细胞仪测定技术研究环孢菌素 A(Cs A)、他莫西芬 (TAM)及α-干扰素 (IFN-α)单独应用和半量联合应用逆转裸鼠高致瘤性多药耐药人白血病细胞系 K5 6 2 - n/ VCR的作用。结果 :Cs A、TAM、IFN -α 3种药物单独和联合应用对 K 5 6 2 - n/ VCR细胞系的多药耐药均有逆转作用 ,Cs A单用逆转效果远远强于TAM及 IFN -α,Cs A>IFN -α>TAM;两两半量联合效果视不同组合而不同 ,三者半量联合应用比两两半量联合逆转效果强 ,而两两半量联合及三者半量联合应用的逆转效果与单独应用 Cs A相似 ,优于单独应用 TAM和 IFN-α。 Cs A使多药耐药细胞系 K 5 6 2 - n/ VCR细胞内 DNR含量增加 ,TAM及IFN-α不改变 K5 6 2 - n/ VCR细胞内 DNR含量。结论 :Cs A、TAM及 IFN -α 3种药物单独和联合应用对 K5 6 2 - n/ VCR细胞的多药耐药均有逆转作用 ,联合应用逆转作用比单独作用强 ,将 Cs A+TAM两药联合或 Cs A+TAM+IFN-α三药联合与化疗药物同时应用可能更有利于提高 MDR白血病患者的治愈率
Objective: To find a suitable method to reverse the multidrug resistance in clinical practice. The combination of the three doses was better than the combined effect of two and a half doses, The reversal effect was similar to that of Cs A alone, which was superior to TAM and IFN-α alone. CsA increased the content of DNR in K562-n / VCR cells, while TAM and IFN-α did not change the content of DNR in K562-n / VCR cells. CONCLUSION: CsA, TAM and IFN-α alone or in combination can reverse the multidrug resistance of K5 6 2 - n / VCR cells.
