目的探讨骨桥蛋白(OPN)在动情周期和妊娠早期小鼠输卵管的表达。方法 70只昆明雌性小白鼠分为动情周期组、妊娠组和假孕组,昆明雄性小白鼠15只用于与雌鼠合笼。取雌鼠动情周期、妊娠和假孕第1~5天的输卵管,利用免疫组织化学染色及半定量分析检测OPN在小鼠输卵管的定位和表达情况。结果整个动情周期小鼠输卵管上皮组织OPN的免疫染色均呈阳性,动情期的输卵管上皮OPN染色的平均吸光度值(AA)最高,动情间期组AA值最低;妊娠和假孕第1~5天小鼠输卵管上皮OPN的免疫染色均为阳性。妊娠第4天输卵管上皮OPN染色的AA值最高;妊娠组第3~5天OPN染色的AA值显著高于假孕组所对应的妊娠天数(P<0.05,P<0.01,P<0.05)。结论小鼠动情周期输卵管上皮OPN的表达可能受体内雌激素的调节,妊娠早期小鼠输卵管上皮OPN的表达可能受雌激素和着床前胚胎共同调节。
Objective To investigate the oviduct expression of osteopontin (OPN) in estrous cycle and early pregnancy. Methods Seventy Kunming female mice were divided into estrous cycle group, pregnancy group and pseudopregnancy group. Fifteen Kunming male mice were used for cages with female rats. Take estrous cycle female estrus, pregnancy and pseudopregnancy 1 to 5 days of fallopian tubes, the use of immunohistochemical staining and semi-quantitative analysis OPN in mouse oviduct positioning and expression. Results Immunostaining of OPN in the oviduct epithelial tissue of mice during the entire estrous cycle was positive. The average absorbance value (OPN) of oviduct epithelial OPN in estrus was the highest, and the AA value was lowest in estrous period group. The first to fifth day of pregnancy and pseudopregnancy Mouse oviduct epithelial OPN immunostaining were positive. The AA value of OPN staining in the fallopian tube epithelium was the highest on the 4th day of gestation. The AA value of OPN staining in the 3rd to 5th day of gestation group was significantly higher than that of the pregnancy group (P <0.05, P <0.01, P <0.05). Conclusion The oval epithelial OPN expression in mouse estrous cycle may be regulated by estrogen in vivo. The expression of OPN in oviduct epithelium in early pregnancy may be regulated by estrogen and preimplantation embryo.
