目的:探讨中药鳖甲煎丸药物血清对人肾小球系膜细胞(MCs)增殖和凋亡作用的影响。方法:应用细胞培养技术,进行人MCs培养,采取血清药理学方法,制备鳖甲煎丸药物血清,利用脂多糖(LPS)为刺激因子,将培养的MCs分为空白对照组(简称“空白组”);鳖甲煎组;脂多糖组;脂多糖鳖甲煎组(简称“脂甲组”)。应用四甲基偶氮唑蓝(MTT)法和流式细胞仪检测MCs增殖、凋亡和细胞周期。结果:LPS刺激后,增殖指数明显提高,与空白组比较有明显差异(P<0.01);鳖甲煎丸药物血清能明显抑制人MCs增殖(P<0.01),并且使多数MCs阻滞在G0/G1期,能诱导人MCs发生凋亡。结论:抑制MCs增殖和促进凋亡可能是鳖甲煎丸延缓慢性肾脏疾病进展的部分作用机制。
OBJECTIVE: To investigate the effect of serum of Biejiajian Pill on proliferation and apoptosis of human mesangial cells (MCs). METHODS: Cell culture techniques were used to culture human MCs. Serum pharmacological methods were used to prepare the serum of Biejiajian Pills. Lipopolysaccharide (LPS) was used as the stimulatory factor. Cultured MCs were divided into blank control groups (abbreviated as “blank”). Group “); armor decoction group; lipopolysaccharide group; lipopolysaccharide armor decoction group (abbreviated ”fat armor group"). The proliferation, apoptosis and cell cycle of MCs were detected by MTT assay and flow cytometry. Results: After LPS stimulation, the proliferation index increased significantly compared with the blank group (P<0.01); Biejiajian Pill serum could significantly inhibit the proliferation of human MCs (P<0.01), and most of the MCs were arrested at G0. /G1 phase can induce apoptosis of human MCs. Conclusion: Inhibiting MCs proliferation and promoting apoptosis may be part of the mechanism by which Biejiajian Pills delay the progression of chronic kidney disease.
