目的:观察银杏叶提取物(GbE)对肝纤维大鼠肝组织iNOS、ET表达的影响,探讨GbE抗肝纤维的作用机制。方法:采用40%的CCl4皮下注射,3 mL.kg-1.d-1,2次/周,首剂加倍,连续6周诱导大鼠肝纤维化模型,同时分别以高、中、低剂量(200、100、50 mg.kg-1.d-1)的GbE灌胃干预6周,RT-PCR及免疫组化法分别检测肝组织iNOS和ET基因的mRNA及蛋白表达。结果:模型组大鼠肝组织iNOS和ET基因的mRNA及蛋白表达均较正常组明显增高;应用高、中、低剂量GbE干预后,大鼠肝组织iNOS和ET基因的mRNA及蛋白表达明显下降。结论:GbE下调肝纤维化大鼠肝组织iNOS和ET基因的转录和表达可能是其抗肝纤维化作用的机制之一。
Objective: To observe the effect of Ginkgo biloba extract (GbE) on the expression of iNOS and ET in hepatic tissue of hepatic fibrosis rats and to explore the mechanism of action of GbE on hepatic fibrosis. Methods: 40% CCl4 subcutaneous injection, 3 mL.kg-1.d-1, 2 times / week, the first dose doubled for 6 weeks to induce rat liver fibrosis model, respectively, at high, medium and low dose (200,100,50 mg.kg-1.d-1) for 6 weeks. The mRNA and protein expression of iNOS and ET in liver tissues were detected by RT-PCR and immunohistochemistry respectively. Results: The mRNA and protein expressions of iNOS and ET in liver of model group were significantly higher than those in normal group. The mRNA and protein expression of iNOS and ET in liver decreased significantly after high, medium and low dose GbE intervention . Conclusion: Down-regulation of iNOS and ET gene transcription and expression by hepatic fibrosis in GbE may be one of the mechanisms of anti-hepatic fibrosis.
