为了进一步探讨 p CDSj32诱发小鼠抗血吸虫感染的保护免疫机制 ,分别用 10 0 μg和 30 0 μg p CDSj32质粒免疫小鼠 ,观察血清 Ig G抗体滴度的动态变化。结果显示 ,单剂量免疫后 4wk,血清 Ig G滴度可达 1∶ 32 0以上 ,以后血清抗体滴度均维持在 1∶ 6 40~ 12 80 ,最少可持续 2 4wk。末次取血清后 ,用日本血吸虫尾蚴攻击感染后 7d,测定血清抗体滴度可骤升至 1∶ 2 5 6 0以上。抗体依赖细胞介导的细胞毒试验表明 ,10 0 μg p CDSj32质粒免疫后 4wk及 8wk血清可介导小鼠腹腔巨噬细胞产生很强的体外杀伤日本血吸虫童虫作用
In order to further explore the protective immunity mechanism of pCDSj32-induced anti-schistosome infection in mice, mice were immunized with 100 μg and 30 μg of CDSj32 plasmid respectively to observe the dynamic changes of serum Ig G antibody titers. The results showed that 4wk after a single dose of serum Ig G titers up to 1: 32 0 above, after the serum antibody titers were maintained at 1: 640 ~ 1280, at least sustainable 24wk. After the last serum was taken, with the Schistosoma japonicum cercariae infection 7d after infection, the determination of serum antibody titer can be increased to 1: 2560 or more. Antibody-dependent cell-mediated cytotoxicity assays showed that 4wk and 8wk sera of 100 μg pCDSj32 plasmid could induce peritoneal macrophages in mice to produce strong killing effects on Schistosoma japonicum
