目的:探讨在大鼠脑缺血/再灌注脑水肿中水通道蛋白-4(AQP4)表达水平的动态变化与脑含水量之间的关系。方法:50只SD大鼠,随机分为假手术组和6,12,24,48 h脑缺血/再灌注模型组,Western blot法测定AQP4的表达,干湿重法测定脑含水量。结果:与假手术组比较,模型组大鼠缺血侧脑含水量和AQP4的表达水平随着时间的推移不断增加,均在24 h时达到最高峰;12~24 h时间段脑含水量显著增加(P<0.01),以及AQP4表达水平升高,48 h时两者均稍有下降。而健侧脑含水量和APQ4的表达水平各时间点之间差异不显著。采用Spearman相关性分析结果显示,脑含水量和AQP4表达水平相关性显著(P<0.05)。结论:大鼠脑缺血/再灌注后脑含水量和AQP4表达水平变化趋势基本一致,两者呈时相正相关性。
AIM: To investigate the relationship between dynamic changes of aquaporin-4 (AQP4) expression and brain water content in rats with cerebral edema after cerebral ischemia / reperfusion. Methods: Fifty Sprague-Dawley rats were randomly divided into sham operation group and 6, 12, 24 and 48 h ischemia / reperfusion models. The expression of AQP4 was detected by Western blot and the brain water content was measured by wet and dry method. Results: Compared with the sham-operation group, the content of water and the expression of AQP4 in the ischemic brain of model group increased gradually with the passage of time, both reaching the peak at 24 h and the brain water content at 12 ~ 24 h (P <0.01), and the expression of AQP4 increased, both of which decreased slightly at 48 h. The contralateral brain water content and APQ4 expression levels between the time points was not significantly different. Spearman correlation analysis showed that the correlation between brain water content and AQP4 expression was significant (P <0.05). CONCLUSION: The changes of brain water content and AQP4 expression level after cerebral ischemia / reperfusion in rats are basically the same, the positive correlation between them is in time phase.
