目的确定人 IgA Fc受体( Fcα R, CD89)结合 IgA的功能部位。方法先采用 PCR及点突变 PCR技术获得不同片段的 Fcα R cDNA,再将各片段分别插入表达质粒 pET30a中,在大肠杆菌中表达出不同片段的受体,用一组能够结合 Fcα R的单克隆抗体通过 Western印迹法鉴别所表达片段是否具有 Fcα R的功能部位。结果表达了 5种不同片段的 Fcα R,能够封闭 Fcα R的中和抗体仅与受体的远膜区( EC1)结合。结论 Fcα R的功能部位位于 EC1上。
Objective To determine the functional site of human IgA Fc receptor (Fcα R, CD89) that binds to IgA. Methods The Fcα R cDNA of different fragments was obtained by PCR and point mutation PCR. The fragments were respectively inserted into the expression plasmid pET30a. The expressed fragments of different receptors were expressed in Escherichia coli. Using a set of monoclonal antibodies capable of binding to FcαR, Antibodies were identified by Western blotting to determine if the expressed fragment possesses a functional site for FcαR. As a result, FcαR of five different fragments was expressed, and the neutralizing antibody capable of blocking FcαR bound only to the far membrane region (EC1) of the receptor. Conclusion The functional site of Fcα R is located on EC1.
