目的建立校正紫外分光光度法测定鹰嘴豆总异黄酮含量的方法。方法采用紫外分光光度法,以染料木素(Genistein)为标准品,测定不同比例染料木素和芒柄花素混合标准液的总异黄酮浓度以及校正前后样品的回收率,检测波长262nm和250nm。结果混合标准液中不同异黄酮含量的比例严重影响测定精确度(测定值/实际值),回归分析显示测定精确度与混合标准液在262nm和250nm处的吸光度之比呈显著线性相关,通过该相关性可校正计算混合溶液异黄酮测定值,加样回收试验表明校正前后的平均回收率分别为93.74%和97.95%。结论该校正方法简单、可靠,可显著提高鹰嘴豆总异黄酮的测定精确度。
Objective To establish a method for the determination of total isoflavone content in chickpea by UV spectrophotometry. Methods Genistein was used as a standard to determine the total isoflavone concentration of different ratios of genistein and formononetin mixed standard solution and the recoveries of samples before and after calibration by using ultraviolet spectrophotometry. The detection wavelength was at 262nm and 250nm . Results The ratio of different isoflavone content in mixed standard solution seriously affected the determination accuracy (measured value / actual value). Regression analysis showed that the determination accuracy was significantly linearly correlated with the absorbance ratio of mixed standard solution at 262nm and 250nm. Correlation Calculate the measured value of isoflavone in mixed solution. The average recovery of sample before and after calibration shows 93.74% and 97.95% respectively. Conclusion The calibration method is simple and reliable, which can significantly improve the determination accuracy of total isoflavones in chickpea.
