目的:研究疏血通对脑缺血大鼠细胞凋亡的影响。方法:雄性Wistar大鼠72只,随机分成脑缺血组、脑缺血+疏血通治疗组、假手术对照组,每组24只,采用大脑中动脉栓塞方法,制作脑缺血模型,疏血通组腹腔注射疏血通注射液0.2ml/kg,脑缺血组及假手术对照组腹腔注射等体积生理盐水。用末端脱氧核苷酸转移酶介导的缺口末端标记法(TUNEL)检测脑组织中神经细胞凋亡。结果:脑缺血组TUNEL染色阳性细胞数显著增多,并随时间的延长而加重,第3天达高峰。脑缺血+疏血通治疗组各时间点与脑缺血组比较TUNEL染色阳性细胞数明显减少(P<0.01)。结论:疏血通能够明显抑制脑缺血大鼠神经细胞凋亡。
Objective: To study the effect of Shuxuetong on apoptosis of rats with cerebral ischemia. METHODS: Seventy-two male Wistar rats were randomly divided into cerebral ischemia group, cerebral ischemia + Shuxuetong group, and sham-operated control group. 24 rats in each group were treated with middle cerebral artery embolization to make cerebral ischemia model. Xuetong Injection was injected intraperitoneally with 0.2 ml/kg Shuxuetong injection. The cerebral ischemia group and sham-operated control group were injected with an equal volume of normal saline. Apoptosis of nerve cells in brain tissue was detected by terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL). RESULTS: The number of TUNEL-positive cells in the cerebral ischemia group increased significantly and increased with time, peaking on the third day. Compared with the cerebral ischemia group, the number of TUNEL positive cells in the cerebral ischemia + Shuxuetong group was significantly decreased (P<0.01). Conclusion: Shuxuetong can obviously inhibit neuronal apoptosis in rats with cerebral ischemia.
