目的探讨黄连素(Ber)对实验性自身免疫性脑脊髓炎(EAE)大鼠脊髓内基质金属蛋白酶-9(MMP-9)、细胞间黏附分子-1(ICAM-1)的影响。方法Lewis大鼠24只随机分为3组:正常组、模型组和实验组。用行为学观察发病情况,常规HE染色观察中枢神经系统的病理变化;用免疫组化法和实时聚合酶链反应(RT-PCR)法,检测大鼠脊髓MMP-9、ICAM-1的变化。结果与模型组比较,实验组神经症状和病理改变减轻,脊髓内MMP-9和ICAM-1的蛋白及mRNA的表达均明显下降(P<0.05)。结论黄连素对EAE大鼠有神经保护作用,其可能机制与下调MMP-9和ICAM-1的蛋白和基因水平表达有关。
Objective To investigate the effects of berberine on the expression of matrix metalloproteinase-9 (MMP-9) and intercellular adhesion molecule-1 (ICAM-1) in spinal cord of experimental autoimmune encephalomyelitis (EAE) rats. Methods Twenty-four Lewis rats were randomly divided into three groups: normal group, model group and experimental group. Pathological changes of the central nervous system were observed by routine HE staining. The changes of MMP-9 and ICAM-1 in the spinal cord were detected by immunohistochemistry and real-time polymerase chain reaction (RT-PCR). Results Compared with the model group, the neurological symptoms and pathological changes in the experimental group were alleviated. The protein and mRNA expressions of MMP-9 and ICAM-1 in the spinal cord were significantly decreased (P <0.05). Conclusion Berberine has neuroprotective effects on EAE rats, and its possible mechanism is related to down-regulation of protein and gene expression of MMP-9 and ICAM-1.
