目的探讨线粒体基因3243和3426位点碱基变异与老年2型糖尿病的易感性。方法采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法,对186例老年2型糖尿病患者和170例健康对照者的线粒体基因3243和3426位点进行筛选分析,并以DNA测序方法确认。结果两组均未检出3243 A→G变异和3426 A→G变异,测序中发现糖尿病组3290 T→C变异1例。结论线粒体基因3243 A→G变异和3426 A→G变异可能与湖北地区老年2型糖尿病的易感性无关联性。
Objective To investigate the susceptibility of mitochondrial DNA polymorphisms at nucleotide positions 3243 and 3426 to type 2 diabetes in the elderly. Methods PCR-RFLP was used to screen and analyze the mitochondrial DNA 3243 and 3426 loci in 186 elderly patients with type 2 diabetes mellitus and 170 healthy controls. Confirmation of sequencing method. Results No mutation of 3243 A → G and 3426 A → G were detected in the two groups. One case of 3290 T → C mutation was found in the diabetic group. Conclusion The variation of mitochondrial gene 3243 A → G and 3426 A → G may not be associated with the susceptibility to type 2 diabetes in the elderly in Hubei province.
