目的通过观察左归丸含药血清对成骨细胞白细胞介素-1(IL-1)、白细胞介素-6(IL-6)和环加氧酶-2(COX-2)表达的影响,探讨其治疗骨质疏松症的作用机制。方法体外分离、培养成骨细胞,实验分为3组:正常血清组、卵巢切除(OVX)血清组和OVX含药血清组。采用免疫组化法,检测成骨细胞IL-1、IL-6和COX-2的表达。结果OVX血清组成骨细胞IL-1、IL-6和COX-2的表达明显强于正常血清组,而OVX含药血清组的表达较OVX血清组明显减弱,与正常血清组比较,则无显著性差异。结论在去势状态下,左归丸可能是通过抑制成骨细胞IL-1、IL-6和前列腺素E2(PGE2)的分泌,进而达到治疗骨质疏松的作用。
Objective To observe the effect of Zuogui Pill-containing serum on the expression of interleukin-1 (IL-1), interleukin-6 (IL-6) and cyclooxygenase-2 (COX-2) in osteoblasts. Explore its mechanism of action for the treatment of osteoporosis. Methods Osteoblasts were isolated and cultured in vitro. The experiments were divided into 3 groups: normal serum group, OVX serum group and OVX drug-containing serum group. Immunohistochemistry was used to detect the expression of IL-1, IL-6 and COX-2 in osteoblasts. RESULTS: The expression of IL-1, IL-6 and COX-2 in OVX serum was significantly stronger than that in normal sera, while the expression in OVX-containing sera was significantly lower than that in OVX sera, but no significant difference was found between OVX serum and normal sera. Sexual differences. Conclusion In the ovariectomized state, Zuogui Pill may be used to treat osteoporosis by inhibiting the secretion of IL-1, IL-6 and prostaglandin E2 (PGE2) from osteoblasts.
