目的通过对两种不同产地大叶钩藤(Uncaria macrophylla Wall.)的核糖体内转录间隔区(rDNA ITS)序列进行RFLP遗传分析,研究不同地理区域大叶钩藤的rDNA ITS序列是否具有生物地理上的差异。方法通过CTAB法提取样品DNA,利用通用引物对其rDNA ITS进行PCR扩增,从而对连接转化后扩增得到的rDNA ITS序列进行限制性片段长度多态性(Restriction Fragment Length Polymorphism,RFLP)分析。结果获得采集自广东肇庆鼎湖山和广西植物园的两种大叶钩藤两种限制性内切酶(MspⅠ&HaeⅢ)酶切图谱。结论基于对rDNA ITS序列进行的RFLP分析,表明这两个地区的大叶钩藤rDNA ITS序列并无明显差异,不同地理区域的大叶钩藤其遗传本质不具有地理差异。
OBJECTIVE: To determine whether the rDNA ITS sequence of Uncaria macrophylla Wall from different geographical regions is biologically geographically analyzed by RFLP based on RFLP analysis. The difference. Methods DNA samples were extracted by CTAB method and their rDNA ITS was amplified by universal primers. Restriction Fragment Length Polymorphism (RFLP) analysis of rDNA ITS sequences amplified by ligation and transformation was carried out. Results Two restriction enzyme (MspⅠ & HaeⅢ) digestion maps were collected from Dinghushan, Zhaoqing, Guangdong and Guangxi Botanical Garden. Conclusion Based on the RFLP analysis of rDNA ITS sequences, there is no significant difference in the rDNA ITS sequences between the two regions. The genetic characteristics of L. glutamicum in different geographical regions have no geographical differences.
