目的:研究pH敏感性壳聚糖海藻酸钠水凝胶体系的溶胀特性及在药物缓释中的应用。方法:在温和条件下制备壳聚糖-海藻酸钠复合水凝胶颗粒,研究其在胃的pH环境(pH=1.4)和肠道pH环境(pH=7.4)的溶胀动力学并考察各种因素对凝胶溶胀性能的影响,利用该凝胶的特性制备酮洛芬微囊,测定其在人工胃液和人工肠液中的缓释效果。结果:该凝胶颗粒在pH=1.4的盐酸缓冲溶液中溶胀度较小,而在pH=7.4的磷酸盐缓冲溶液中溶胀度很大,用它制备的酮洛芬微囊具有明显的缓释效果。结论:讨论了壳聚糖浓度,海藻酸钠浓度,钙离子浓度对凝胶溶胀性能的显著影响以及利用这些特性制备的酮洛芬微囊的缓释效果。
Objective: To study the swelling properties of pH-sensitive chitosan-alginate hydrogel system and its application in drug delivery. METHODS: Chitosan-sodium alginate hydrogel particles were prepared under mild conditions and their swelling kinetics were studied in the pH environment of the stomach (pH = 1.4) and the pH environment of the intestinal tract (pH = 7.4) Factors on the swelling properties of the gel, the use of the characteristics of the gel preparation of ketoprofen microcapsules, measured in the artificial gastric juice and artificial intestinal fluid sustained release effect. Results: The swelling degree of the gel particles in the hydrochloric acid buffer solution with pH = 1.4 was small, while the swelling degree in the phosphate buffer solution with pH = 7.4 was great. The ketoprofen microcapsules prepared by the gel particles had obvious sustained release effect. Conclusion: The effects of chitosan concentration, sodium alginate concentration and calcium ion concentration on the swelling properties of gel were discussed. The sustained-release effect of ketoprofen microcapsules prepared by these properties was also discussed.
