目的:为大黄泻下有效成分在体内的代谢研究提供基础研究资料。方法:大鼠口服给予大黄提取物后,依一定的时间间隔,自颈静脉采血,分离血浆,应用HPLC/MS联用技术测定其中Rhein的含量,绘制药-时曲线。结果:口服给药后,0~6 h内,Rhein在血浆中均有分布,给药后0. 5 h达到峰值。结论:Rhein血药浓度变化,可能对与其结构相似的大黄蒽醌类成分体内代谢的研究具有指导意义。
Objective: To provide basic research data for the metabolism of effective components of rhubarb diarrhea in vivo. METHODS: Rats were orally given rhubarb extract, and blood was collected from the jugular vein at regular intervals. The plasma was separated. The content of Rhein was determined by HPLC/MS and the drug-time curve was plotted. RESULTS: After oral administration, Rhein was distributed in the plasma within 0 to 6 h, peaking at 0.5 h after administration. Conclusion: The change of blood concentration of Rhein may be of significance for the study of in vivo metabolism of rheum astragalus components with similar structure.
