应用放射配基结合分析法(RBA)检测了大鼠不同发育阶段卵巢内皮素-1(ET-1)受体的动态变化。结果表明:大鼠卵巢上存在有特异性ET-1受体,受体最大结合量(Bmax)于窦前期卵泡含量最高(358.2±73.2fmol/mg蛋白),随卵泡发育到排卵前卵泡时显著降低(162.7±17.2fmol/mg蛋白)(P<0.01),排卵后的早期黄体化卵巢降至最低值(37.8±3.4fmol/mg蛋白)(P<0.01),于晚期黄体化卵巢又再次升高(95.3±4.9fmol/mg蛋白)(P<0.01)。受体亲和力(Kd值)也随卵泡和黄体发育的不同阶段呈现动态变化,在排卵前卵泡卵巢ET受体对ET-1的亲和力最低,晚期黄体化卵巢最高。结果提示,ET-1可能通过与ET-1受体结合参与卵泡和黄体发育的调节。本结果为ET-1作为卵巢内局部调节肽提供实验依据。
Radioligand binding assay (RBA) was used to detect the dynamic changes of endothelin-1 (ET-1) receptor in rats at different developmental stages. The results showed that there was a specific ET-1 receptor on the ovary of ovary, and the maximum amount of receptor (Bmax) in preantral follicles was the highest (358.2 ± 73.2 fmol / mg protein) Follicles significantly decreased (162.7 ± 17.2fmol / mg protein) (P <0.01), and the rate of ovariectomized early ovariectomized ovaries decreased to the lowest (37.8 ± 3.4fmol / mg protein) (P < 0.01). The level of luteinized ovary increased again (95.3 ± 4.9fmol / mg protein) (P <0.01). Receptor affinity (Kd) also showed dynamic changes at different stages of follicular and corpus luteum development. Before ovulation, the ovarian ET receptor had the lowest affinity to ET-1 and the late stage luteinized ovary was the highest. The results suggest that ET-1 may participate in the regulation of follicular and corpus luteum development by binding with ET-1 receptor. This result provides an experimental basis for ET-1 as a local regulatory peptide in the ovary.
