目的通过测定急性肺损伤(acute lung injury,ALI)血浆及肺泡灌洗液中尾加压素Ⅱ(urotensinⅡ,UⅡ)的含量,来探讨尾加压素受体拮抗剂(UⅡreceptor antagonist,URA)对ALI中UⅡ的影响。方法Sprague Dawley(SD)大鼠28只,随机分为4组,每组7只:生理盐水对照组(A组);另三组以URA干预油酸型ALI动物模型,分别编为B、C、D组。3h后4个组均抽动脉血作血气分析,A组、B组抽动脉血后随即经心脏取血各2ml待测,C组、D组分别于12h、24h各取同量心脏血待测。各组均于心脏取血后活杀并取肺,以右肺作支气管肺泡灌洗(bronchoalveolar lavage,BAL),灌洗液(bronchoalveolar lavagefluid,BALF)离心后留取上清液,与心脏血分离的血浆一起在-80℃冻存待测。结果血气分析显示,B、C、D组血氧分压明显低于A组。A、B、C、D组血浆UⅡ质量浓度分别为(118·1±9·8)pg/ml、(133·6±8·9)pg/ml、(132·0±9·8)pg/ml、(114·0±9·1)pg/ml;相应各组BALF中UⅡ质量浓度分别为(10·1±1·4)pg/ml、(21·5±3·5)pg/ml、(63·1±6·5)pg/ml、(79·0±8·8)pg/ml。ALI大鼠血浆中UⅡ浓度进行性降低,而BALF中UⅡ进行性升高,差异具有统计学意义。结论UⅡ可能通过活化URA参与ALI的发病过程,并可能对ALI具有某种保护作用。

The concentrations of UⅡin the BALF of the corresponding groups were (10.1 ± 1.4) pg / ml and (21.5 ± 3.5) pg / ml, (63.1 ± 6.5) pg / ml and (79.0 ± 8.8) pg / ml, respectively. The concentration of UⅡ in plasma of ALI rats decreased progressively, while UⅡ in BALF increased progressively, the difference was statistically significant. Conclusion UⅡ may participate in the pathogenesis of ALI through activation of URA and may have some protective effect on ALI.
