[目的]对葡萄渣提取液进行澄清,并对工艺参数进行优化。[方法]选择果胶和纤维素复合酶对葡萄籽提取物进行澄清,分别在最适温度和常温作用、最适pH和不调pH条件下比较了复合酶对葡萄籽提取液澄清效果及多酚和原花青素含量的影响。[结果]试验表明,复合酶对葡萄籽提取液澄清效果受温度的影响较大,调节pH对澄清度的影响则表现不明显。正交试验证明,最佳澄清工艺条件为pH 5.5,酶解温度45℃、作用时间6 h、酶剂量60 mg/kg,透光率达到76%,澄清过程中不影响葡萄籽多酚以及原花青素含量。[结论]研究可为葡萄籽提取液的开发利用提供理论依据。
[Objective] The research aimed to clarify the grape residue extract and optimize the process parameters. [Method] The pectin and cellulose complex enzyme were selected to clarify the grape seed extract. The clarification effect of the composite enzyme on the grape seed extract was compared under the optimum temperature and normal temperature, the optimum pH and the no pH Effect of phenol and proanthocyanidin content. [Result] The results showed that the clarification effect of the compound enzyme on the grape seed extract was greatly affected by the temperature, and the effect of adjusting the pH on the clarity was not obvious. The results of orthogonal experiment showed that the optimum conditions were pH5.5, temperature 45 ℃, reaction time 6 h, enzyme dosage 60 mg / kg and transmittance of 76%. The clarification process did not affect grape seed polyphenols and proanthocyanidins content. [Conclusion] The research could provide a theoretical basis for the exploitation and utilization of grape seed extract.
