建立灵敏、快速的反式阿魏酸(trans-FA)血浆浓度的高效液相色谱-串联质谱(HPLC-MS/MS)分析方法,研究反式阿魏酸在健康中国受试者体内的药动学特征。血浆样品经乙酸乙酯液-液萃取,采用Hedera ODS-2色谱柱分离,流动相为甲醇-5 mmol·L-1乙酸铵水溶液(含0.05%乙酸)(34∶66),流速为0.4 m L·min-1;质谱采用Turbo-Ionspray电喷雾离子源和负离子多反应监测方式。反式阿魏酸在0.1~5 ng·m L-1内线性关系良好(r≥0.999 2);方法的日间和日内精密度均小于9.2%,准确度为95.4%~111.4%;无基质效应,无残留效应;稳定性良好。所建立的HPLC-MS/MS分析方法快速、灵敏、准确、专属性强、重现性高,适用于健康中国受试者体内反式阿魏酸的药代动力学研究。
To establish a sensitive and rapid method for the determination of trans-FA plasma concentration by HPLC-MS / MS, the effect of trans-ferulic acid on healthy Chinese subjects Dynamic characteristics. The plasma samples were separated by liquid-liquid extraction with ethyl acetate and separated on a Hedera ODS-2 column. The mobile phase consisted of methanol-5 mmol·L-1 ammonium acetate (0.05% acetic acid) L · min-1; mass spectrometry using Turbo-Ionspray electrospray ionization source and anion multiple reaction monitoring. The trans-ferulic acid showed a good linearity (r≥0.9992) within 0.1-5 ng · m L-1. The intra-day and intra-day precision was less than 9.2% and the accuracy was 95.4% -111.4% Effect, no residual effect; good stability. The established HPLC-MS / MS method is rapid, sensitive, accurate, specific and reproducible and is suitable for the pharmacokinetic study of trans-ferulic acid in healthy Chinese subjects.
