目的:研究粗壮女贞(Ligustrum robustum BL)对2型糖尿病小鼠的降血糖机制。方法:采用腹腔注射四氧嘧啶(200 mg·kg-1)建立小鼠2型糖尿病模型,实验分为正常对照组、阳性对照组、高、中、低剂量组(24.0g·kg-1·d-1、12.0 g·kg-1·d-1、8.0 g·kg-1·d-1),ELISA试剂盒检测2型糖尿病小鼠血清中血糖值水平(GLU)及血清胰岛素水平,Western blotting法检测小鼠脂肪组织中胰岛素受体(IR-β)、蛋白激酶B(PKB)和葡萄糖转运蛋白4(Glut4)蛋白表达。结果:粗壮女贞提取物能显著降低2型糖尿病小鼠血糖水平(GLU),增加血清胰岛素含量(P<0.05,P<0.01),促进脂肪中胰岛素受体(IR-β)、蛋白激酶B(PKB)和葡萄糖转运蛋白4(Glut4)蛋白的表达。结论:粗壮女贞提取物对2型糖尿病小鼠具有显著的降血糖作用,可显著改善2型糖尿病小鼠糖脂代谢并抑制胰岛素抵抗。
AIM: To investigate the hypoglycemic mechanism of Ligustrum robustum BL in type 2 diabetic mice. Methods: The model of type 2 diabetes in mice was established by intraperitoneal injection of alloxan (200 mg · kg-1). The experiment was divided into normal control group, positive control group, high, medium and low dose groups (24.0g · kg-1 · d-1, 12.0 g · kg-1 · d-1 and 8.0 g · kg-1 · d-1). Serum glucose and serum insulin levels were measured by ELISA kit in type 2 diabetic mice. The protein expression of insulin receptor (IR-β), protein kinase B (PKB) and glucose transporter 4 (Glut4) in mouse adipose tissue was detected by Western blotting. Results: The Fructus Lagbifolia extract could significantly decrease the level of blood glucose (GLU), increase the level of serum insulin (P <0.05, P <0.01), promote the expression of insulin receptor (IR-β), protein kinase B (PKB) and glucose transporter 4 (Glut4) protein expression. Conclusion: Prunella vulgaris extract has a significant hypoglycemic effect on type 2 diabetic mice, which can significantly improve glucose and lipid metabolism and inhibit insulin resistance in type 2 diabetic mice.
