目的探讨miR-21和miR-93的表达水平与三阴性乳腺癌(TNBC)发生发展的相关性。方法选择行乳腺癌根治术的女性患者86例,收集癌组织与正常乳腺组织,采用逆转录聚合酶链反应(RT-PCR)检测miR-21和miR-93的相对表达量,并分析其与临床病理特征的相关性。结果 miR-21和miR-93在TNBC组织中相对表达量高于癌旁组织;TNBC组织中,miR21高表达54例,占62.79%,miR-93高表达50例,占58.14%;miR-21和miR-93在肿瘤直径>5 cm、临床分期为Ⅲ~Ⅳ期以及淋巴结转移的患者中高表达率显著增高(P<0.01);miR-21和miR-93的表达水平与患者年龄、月经状态、病理分级无明显相关性。癌组织中miR-21与miR-93表达水平呈显著正相关(r=0.761,P<0.01)。结论 miR-21与miR-93在TNBC中表达异常增高,且与肿瘤直径、临床分期、淋巴结转移有关,可作为TNBC诊断及判断预后的辅助指标。
Objective To investigate the relationship between the expression of miR-21 and miR-93 and the occurrence and development of triple negative breast cancer (TNBC). Methods Totally 86 female patients with radical mastectomy were enrolled in the study. The relative expression of miR-21 and miR-93 was detected by reverse transcription-polymerase chain reaction (RT-PCR) Correlation of clinicopathological features. Results The relative expression level of miR-21 and miR-93 in TNBC tissues was higher than that in adjacent non-cancerous tissues. In TNBC tissues, miR21 was highly expressed in 54 cases (62.79%) and miR-93 was highly expressed in 50 cases (58.14% (P <0.01). The expression of miR-21 and miR-93 were significantly correlated with the age, menstrual status, and the expression of miR-93 in patients with tumor diameter> 5 cm, clinical stage Ⅲ ~ Ⅳ and lymph node metastasis , No significant correlation between pathological grade. There was a significant positive correlation between the expression of miR-21 and miR-93 in cancer tissues (r = 0.761, P <0.01). Conclusions The expression of miR-21 and miR-93 is abnormally elevated in TNBC, which is related to tumor diameter, clinical stage and lymph node metastasis, which can be used as an auxiliary index for TNBC diagnosis and prognosis.
