目的:建立HPLC法同时测定金荞麦中原儿茶酸和(-)-表儿茶素含量的方法。方法:采用反相高效液相色谱法,Thermo ODS-2HYPERSIL(250mm×4.6mm,5μm)色谱柱,流动相为乙腈-0.3%冰乙酸溶液梯度洗脱,检测波长:原儿茶酸为255nm、(-)-表儿茶素为280nm,流速为1.0ml·min-1,柱温30℃。结果:原儿茶酸进样量在0.0444~0.8880μg线性关系良好(r=0.9999),加样回收率(n=6)103.0%,RSD为1.5%;(-)-表儿茶素进样量在0.05355~1.071μg线性关系良好(r=1.0),加样回收率(n=6)98.5%,RSD为1.7%。结论:该方法准确、快速,可为全面评价金荞麦质量提供依据。
Objective: To establish a method for simultaneous determination of protocatechuic acid and (-) - epicatechin in buckwheat by HPLC. Methods: Reversed-phase high performance liquid chromatography (HPLC) was performed on a Thermo ODS-2HYPERSIL (250 mm × 4.6 mm, 5 μm) column using a gradient elution of acetonitrile-0.3% glacial acetic acid. The detection wavelength was: (-) - epicatechin 280nm, the flow rate of 1.0ml · min-1, the column temperature 30 ℃. Results: The calibration curve of protocatechuic acid was linear in the range of 0.0444-0.88880μg (r = 0.9999), the recovery was 103.0% (n = 6) and the RSD was 1.5%. The (-) - epicatechin injection The linear range was 0.05355 ~ 1.071μg (r = 1.0), the recovery was 98.5% (n = 6), and the RSD was 1.7%. Conclusion: The method is accurate and rapid, which can provide a basis for the comprehensive evaluation of the quality of buckwheat.
