目的 探讨脑胶质瘤与PGE2 之间的内在联系,找出消炎痛类药物做为胶质瘤辅助治疗的可能性。方法 对20 例胶质瘤标本进行细胞培养1周,测其上清液中的PEG2含量。对10例胶质瘤细胞培养液中加入不同浓度消炎痛溶液,培养1 周后测其上清液PGE2 的含量。同时对15 例胶质瘤患者血浆中PGE2 的水平进行测定。结果 胶质瘤合成PGE2 的量高于正常脑组织合成PGE2 的2 倍以上(P<0.001)。不同浓度的消炎痛对胶质瘤合成PGE2 均有明显抑制作用(P< 0.001)。胶质瘤患者血浆中PGE2 含量也明显高于正常人(P< 0.005)。结论 脑胶质瘤生长过程中可以合成大量PGE2,并使其宿主血浆中也含有过量的PGE2 ,消炎痛在体外能有效地抑制胶质瘤细胞合成PGE2 ,这提示消炎痛类药物在胶质瘤治疗中有积极意义
Objective To investigate the intrinsic relationship between glioma and PGE2, and to find out the possibility of indomethacin as an adjuvant therapy for glioma. Methods Twenty glioma specimens were cultured for one week and their supernatants were assayed for PEG2 content. Different concentrations of indomethacin solution were added to 10 glioma cell cultures, and the supernatant PGE2 content was measured after 1 week of culture. At the same time, plasma PGE2 levels were measured in 15 glioma patients. Results The amount of PGE2 synthesized by gliomas was more than 2 times higher than that of normal brain tissue (P<0.001). Different concentrations of indomethacin significantly inhibited the synthesis of PGE2 in gliomas (P<0.001). Plasma PGE2 levels in glioma patients were also significantly higher than those in normal controls (P < 0.005). Conclusion In the process of glioma growth, a large amount of PGE2 can be synthesized, and the host plasma also contains excess PGE2. Indomethacin can effectively inhibit the synthesis of PGE2 in glioma cells in vitro, suggesting that indomethacin drugs are present in gliomas Positive in treatment
