循环肿瘤细胞(circulating tumor cells,CTC)是指从实体肿瘤原发或转移病灶脱落并侵入外周血循环的肿瘤细胞,是恶性肿瘤远处转移的主要原因。目前CTC检测技术发展迅速,但由于外周血中CTC个数稀少且存在较大的异质性,限制了许多检测技术的应用。SE-i FISH技术将差相富集、瘤标免疫荧光染色与染色体荧光原位杂交(immunostaining-fluorescence in situ hybridization,i-FISH)结合,不依赖肿瘤上皮细胞表面标示物的表达,对CTC同时进行瘤标染色与i-FISH染色体计数的双重检测,具有高灵敏性和高特异性。现就CTC检测方法及SE-i FISH技术检测CTC的临床应用作一综述。
Circulating tumor cells (CTCs) refer to tumor cells that fall off from the primary or metastatic lesions of solid tumors and invade the peripheral blood circulation, which is the main reason for the distant metastasis of malignant tumors. At present, CTC detection technology is developing rapidly, but due to the small number of CTCs in the peripheral blood and the large heterogeneity, the application of many detection technologies is limited. The SE-i FISH technique combines differential phase enrichment, tumor immunofluorescence staining and ios immunofluorescence in situ hybridization (i-FISH), and does not depend on the expression of tumor cell surface markers. Simultaneously, Tumor staining and i-FISH chromosome count double detection, with high sensitivity and high specificity. The clinical application of CTC detection and SE-i FISH to detect CTC are reviewed.
