目的:检测长链非编码RNA(lncRNA)ANRIL在肾癌及正常组织、细胞中的表达水平,探究其在肾癌中的临床意义。方法:提取肾癌细胞(ACHN、786O和769P)、正常人胚肾细胞(293T)和47对组织中的总RNA,逆转录获取cDNA,之后通过qPCR方法检测细胞及组织中的ANRIL表达量,分析配对组织、细胞系之间的表达差异,探究ANRIL表达水平与患者临床特征之间的关系。结果:ANRIL在肾癌细胞和组织中表达水平明显低于正常肾细胞(P<0.05)和配对癌旁正常组织(P<0.01),标本中共有34例(72.34%)肾癌组织ANRIL表达低于癌旁组织,癌组织中ANRIL表达与患者年龄、性别及肾癌病理类型无明显相关性(P>0.05),与患者TNM分期和AJCC分级相关(P<0.05)。结论:ANRIL在肾癌组织及细胞系中表达明显下调,且和肾癌临床分级相关,提示其在肾癌发生发展中发挥着一定的作用。
OBJECTIVE: To detect the expression of long chain non-coding RNA (ANRIL) in renal cell carcinoma and normal tissues and cells, and to explore its clinical significance in renal cell carcinoma. METHODS: The total RNA was extracted from renal cancer cells (ACHN, 786O and 769P), normal human embryonic kidney (293T) and 47 pairs of tissues. CDNA was reverse transcribed. Then the expression of ANRIL in cells and tissues was detected by qPCR. Analysis of paired tissues, cell lines between the expression differences, explore the ANRIL expression levels and the relationship between the clinical features of patients. Results: The expression of ANRIL in renal cell carcinoma and tissues was significantly lower than that in normal renal cells (P <0.05) and matched adjacent normal tissues (P <0.01). ANRIL expression was low in 34 (72.34% There was no significant correlation between the expression of ANRIL and the age, sex and pathological type of renal cell carcinoma (P> 0.05). The expression of ANRIL was correlated with TNM stage and AJCC grade (P <0.05). Conclusion: The expression of ANRIL in renal cell carcinoma and cell lines was significantly down-regulated and correlated with the clinical grade of renal cell carcinoma, suggesting that it may play a role in the development of renal cell carcinoma.
