锌指核酸酶是一种人工合成的融合蛋白,由特异性识别靶DNA的锌指蛋白结构域连接非特异性的FokI切割结构域构造而成,能够在预定的DNA靶位点引起DNA双链中断,随后依靠非同源末端连接和同源重组修复两种途径修饰该位点.近些年来,该技术已广泛的应用于基因转变、基因修饰和基因治疗等方面的研究.本文将简要介绍锌指核酸酶结构与原理,重点对锌指核酸酶的设计和应用进行了回顾与展望.
