为探讨白细胞介素 -6( IL -6)在卵巢切除诱导的大鼠骨吸收中的作用及补肾中药密骨片对其的影响。将 3 0只 SD大鼠随机分为假手术组 ( Sham ) ,卵巢切除组 ( OV X)及密骨片治疗组 ( OVX-MGP)。分离三组大鼠右股骨骨髓上清 ,以 10 %~ 60 %浓度加入 45Ca标记的胎鼠颅骨培养液中。观察骨吸收活性并通过 EL ISA测定骨髓上清 IL-6含量。结果示骨髓上清 IL-6浓度与骨吸收活性呈量效关系 ;OVX组骨吸收活性较 Sham组显著升高 ,OVX-MGP组较 OVX组明显下降 ( P均 <0 .0 1) ;OVX组骨髓上清 IL-6浓度是 Sham组的 2倍 ,与 OV X比较 ,O VX-MGP组 IL -6含量下降 5 2 % ( P均 <0 .0 1)。表明卵巢切除导致内源性骨吸收因子 IL -6产生增加 ,密骨片抗骨吸收作用部分是通过降低 IL-6产生而介导的。
To investigate the role of interleukin-6 (IL-6) in ovariectomy-induced rat bone resorption and the effect of Bushen Chinese medicine TCM on its bone resorption. Thirty SD rats were randomly divided into sham-operated group (Sham), ovariectomized group (OVX) and Migu Tablet-treated group (OVX-MGP). The right femoral bone marrow supernatant of three groups of rats was isolated and added to the 45Ca-labeled rat calvarial culture fluid at a concentration of 10% to 60%. The bone resorption activity was observed and IL-6 content in the supernatant of the bone marrow was measured by EL ISA. The results showed that there was a dose-effect relationship between the concentration of IL-6 in the bone marrow supernatant and the bone resorption activity; the bone resorption activity in the OVX group was significantly higher than that in the Sham group, and the OVX-MGP group was significantly lower than the OVX group (P<0.01); The concentration of IL-6 in the supernatant of the bone marrow was 2 times that of the Sham group. Compared with the OV X, the IL-6 content in the O VX-MGP group was decreased by 52% (P<0.01). This indicates that ovariectomy results in an increase in the production of the endogenous bone resorption factor IL-6, and that the bone resorption of melastoid slices is mediated in part by decreasing IL-6 production.
