目的:建立同时测定人血清中缬氨酸、亮氨酸、异亮氨酸、苯丙氨酸和酪氨酸的分析方法。方法:以稳定同位素标记的氨基酸作为内标,血清样本经乙腈沉淀蛋白后,取上清液进行HILIC-MS/MS分析,色谱柱为Syncronis HILIC(150 mm×2.1 mm,5μm),流动相为乙腈-80mmol·L-1醋酸铵,梯度洗脱。结果:基质效应不影响测定结果,各氨基酸在标准曲线范围内线性关系良好,准确度、批内和批间精密度均小于15%,在测定过程中稳定性符合要求。结论:该方法可同时测定人血清中5种内源性氨基酸的含量。
Objective: To establish a method for the simultaneous determination of valine, leucine, isoleucine, phenylalanine and tyrosine in human serum. Methods: A stable isotope labeled amino acid was used as an internal standard. Serum samples were precipitated with acetonitrile and the supernatant was subjected to HILIC-MS / MS. The mobile phase consisted of Syncronis HILIC (150 mm × 2.1 mm, 5 μm) Acetonitrile-80mmol · L-1 ammonium acetate, gradient elution. Results: The matrix effects did not affect the determination results. The amino acids showed a good linearity within the standard curve with accuracy less than 15% within and between batches. The stability of the amino acids was within the standard. Conclusion: This method can simultaneously determine the contents of five kinds of endogenous amino acids in human serum.
