目的:探讨生长激素(GH)联合氟尿嘧啶(5-Fu)在体外对结肠癌LOVO细胞的影响。方法:将培养的结肠癌LOVO细胞分为对照组、GH组、5-Fu组和GH+5-Fu组,GH组又按剂量分为两个亚组,即GH1终浓度为50ng/mL和GH2终浓度为100 ng/mL;GH+5-Fu组又按剂量分为两个亚组,即GH1+5-Fu(GH 50 ng/mL+5-Fu 100μg/mL)和GH2+5-Fu(GH 100 ng/mL+5-Fu 100μg/mL),加入不同浓度的处理液后24、48和72 h,分别行四基偶氮唑蓝(MTT)比色观察,计算细胞存活率。并于加入不同浓度的处理液24 h后,以流式细胞仪测定细胞周期和凋亡情况。结果:5-Fu组和GH+5-Fu组与对照组比较,细胞存活率明显下降(P<0.05)。GH+5-Fu组与单用5-Fu组比较,细胞生存率均有所降低,但各时间段无显著性差异(P>0.05)。GH组与对照组比较,S期细胞百分比和增殖指数(PI)均增加(P<0.05),G0/G1细胞百分比有所减少(P<0.05)。GH+5-Fu组的S期细胞百分比和PI均降低(P<0.05)。5-Fu组和GH+5-Fu组的细胞凋亡率均升高(P<0.05);GH+5-Fu组与5-Fu组比较,S期细胞百分比、PI和凋亡率均无显著性差异(P>0.05)。结论:rhGH体外作用于结肠癌LOVO细胞,未刺激细胞生长,GH与化疗联合用是安全的。
Objective: To investigate the effects of growth hormone (GH) combined with 5-fluorouracil on colon cancer LOVO cells in vitro. Methods: The cultured colon cancer LOVO cells were divided into control group, GH group, 5-Fu group and GH + 5-Fu group. The GH group was divided into two subgroups according to the dosage: GH1 final concentration was 50ng / mL and The GH2 + 5-Fu group was subdivided into two subgroups: GH1 + 5-Fu (GH 50 ng / mL + 5-Fu 100 μg / mL) and GH2 + 5- Fu on GH 100 ng / mL + 5-Fu 100 μg / mL. Cell viability was calculated by MTT colorimetric assay 24, 48 and 72 h after the addition of different concentrations of treatment solution. After addition of different concentrations of treatment solution for 24 h, the cell cycle and apoptosis were determined by flow cytometry. Results: Compared with the control group, the cell viability of 5-Fu group and GH + 5-Fu group decreased significantly (P <0.05). The survival rate of GH + 5-Fu group was lower than that of 5-Fu alone group, but there was no significant difference in each time period (P> 0.05). Compared with the control group, the percentage of cells in S phase and the proliferation index (PI) increased (P <0.05) and the percentage of G0 / G1 cells decreased (P <0.05). The percentage of S phase cells and PI in GH + 5-Fu group decreased (P <0.05). 5-Fu group and GH + 5-Fu group (P <0.05). Compared with 5-Fu group, the percentages of cells in S phase, PI and apoptosis rate in GH + 5- Significant difference (P> 0.05). CONCLUSION: rhGH acts on colon cancer LOVO cells in vitro without stimulating cell growth. It is safe to use GH in combination with chemotherapy.
