[目的]研究α-淀粉酶和蛋白酶辅助水提马尾松(Pinus massoniana Lamb.)花粉的工艺参数。[方法]以水提物得率为指标,在单因素试验基础上进一步通过正交试验优化了酶法辅助水提马尾松花粉的工艺,并进行了验证试验。[结果]经过超临界CO2萃取技术脱脂后的马尾松花粉利用α-淀粉酶辅助水提,得到的优化工艺为:加酶量2.0%,料液比1∶16,提取温度75℃,提取时间3h,恒定pH值6.0。在优化后的α-淀粉酶水提松花粉提取物中添加蛋白酶进一步水提,得到的优化工艺为:加酶量1.5%,料液比1∶20,提取温度50℃,提取时间3h,恒定pH值7.0。在优化工艺参数下,水提物的得率为51.34%。[结论]为更加合理和充分地利用松花粉资源提供科学依据。
[Objective] The research aimed to study the process parameters of α-amylase and protease-assisted pollen of Pinus massoniana Lamb. [Method] Taking the rate of water extract as index, the process of enzymatic assisted watering Masson Pine pollen by the orthogonal experiment was further optimized on the basis of single factor experiment, and the validation test was carried out. [Result] The results showed that the optimum technological conditions were as follows: enzyme 2.0%, solid - liquid ratio 1:16, extraction temperature 75 ℃, extraction time 3h, constant pH 6.0. After the optimal aqueous extract of α-amylase was extracted by adding protease, the optimized process was as follows: the amount of enzyme was 1.5%, the solid-liquid ratio was 1:20, the extraction temperature was 50 ℃, the extraction time was 3h, and the constant pH 7.0. Under the optimized process parameters, the yield of water extract was 51.34%. [Conclusion] It provided a scientific basis for more rational and full use of pine pollen resources.
